1/2MS Medium (containing sucrose and agar) is a low-salt derivative of the classic MS medium. It is created by halving the concentrations of macronutrients, micronutrients, and organic components in MS medium (the iron source is usually kept unchanged or slightly adjusted proportionally), while pre-mixing sucrose (carbon source) and agar (solidifying agent) to build a solid culture system with low osmotic pressure and low nutritional strength. This medium balances nutritional adequacy with a gentle environment and is widely used in plant tissue culture for seedling rooting and acclimatization, seed germination, and the cultivation of sensitive materials.
