1/4MS Medium (containing sucrose and agar) is a derivative version of the MS medium family with lower nutrient strength. It reduces the concentrations of macronutrients, micronutrients, and organic components of MS medium to one-quarter of the original formula (the iron source usually remains at the MS level or is slightly adjusted proportionally), and pre-mixes sucrose (carbon source) and agar (gelling agent) to create a solid culture system with extremely low osmotic pressure and nutrient strength. This medium's core advantage lies in providing a "mild nutritional environment," making it irreplaceably valuable in plant tissue culture seedling acclimatization, germination of sensitive seeds, in vitro preservation of rare plants, and studies of low-nutrient stress.
