3/4MS Medium (containing sucrose and agar) is a derivative formula of the MS medium series with moderate nutritional strength. By adjusting the concentrations of macronutrients, micronutrients, and organic components of the MS medium to three-quarters of the original formula (with the iron source usually maintained at the MS level) and pre-mixing sucrose and agar, it creates a solid culture system that balances nutrient supply and environmental adaptability. This medium fills the nutritional gradient gap between MS and 1/2MS, playing a unique role in the 'intermediate stage' of plant tissue culture, such as seedling strengthening, secondary bud induction, and the cultivation of moderately sensitive materials.
