1/8MS Medium (containing sucrose and agar) is a derivative of the MS medium family with extremely low nutrient strength. It is formulated by reducing the concentrations of macronutrients, micronutrients, and organic components of MS medium to 1/8 of the original formula (the iron source is usually maintained at MS level or halved to prevent iron-deficiency chlorosis) and by premixing low concentrations of sucrose and soft agar to create a "ultra-low nutrient, ultra-low osmotic pressure" solid culture system. This medium is designed with the core concept of "simulating naturally infertile environments" and has irreplaceable application value in the final acclimatization of tissue culture seedlings, the germination of extremely sensitive seeds, the in vitro preservation of rare plants, and studies involving extreme low-nutrient stress.
