B5 Medium (containing sucrose and agar) is a classic plant tissue culture medium developed by the Gamborg team in 1968. Its core feature is the inorganic salt composition with a high potassium nitrate content and relatively low ammonium nitrogen, combined with premixed sucrose (as a carbon source) and agar (as a gelling agent), creating a solid culture system suitable for dicotyledons and some monocotyledons. This medium is widely used in plant cell suspension cultures, callus induction, protoplast culture, and genetic transformation of dicotyledons (such as soybean and tobacco), and is particularly suitable for plant materials sensitive to ammonium nitrogen.
