N6 Medium (containing sucrose and agar) is a specialized medium developed in 1975 by the team of Chinese scientist Zhu Zhiqing for the culture of rice anthers. Its core feature is the balance of inorganic salts with high concentrations of potassium nitrate and low ammonium nitrogen, combined with an optimized mix of trace elements, creating a solid system suitable for the in vitro culture of gramineous plants (especially cereal crops such as rice and wheat). This medium is widely used in monocot anther culture, pollen plant induction, young embryo culture, and genetic transformation, and it is particularly noted for its unique ability to promote male gametophyte development in gramineous plants.
