White Medium(with Sucrose& Agar)
White Medium(with Sucrose& Agar)
White Medium (containing sucrose and agar) is a classic plant tissue culture medium developed by White in 1943. Its core feature is the low concentration of inorganic salts (especially nitrogen and potassium, which are much lower than in media such as MS or B5) combined with a simplified nutrient composition. With premixed sucrose (as a carbon source) and agar (as a gelling agent), it establishes a low-nutrient solid system suitable for the cultivation of plant roots, embryos, and sensitive tissues (such as orchid protocorms and fern sporophytes). This medium is widely used in ex vitro root culture, virus detection (e.g., potato virus identification), and long-term preservation of materials with low metabolic demands, and is particularly suitable for plant tissues that are sensitive to high ion concentrations.
