NS Medium (containing sucrose and agar) is a specialized culture medium commonly used for plant protoplast culture and somatic hybridization. Its core feature is a combination of low-osmotic-pressure inorganic salts and optimized organic nutrients. By precisely regulating the concentrations of nitrogen and potassium ions (lower than MS and B5 media) and pre-mixing sucrose (a carbon source) and agar (a gelling agent), it creates a mild solid environment suitable for protoplast survival, cell wall regeneration, and cell colony formation. This medium is widely used in protoplast cultures of crops such as tobacco and tomato, particularly in reducing osmotic stress caused by high ion concentrations on cell wall-free protoplasts, significantly improving protoplast survival rates and subsequent division capabilities.
