NB Medium (containing sucrose and agar) is a plant tissue culture medium improved based on the N6 medium. Its core feature is retaining the high potassium and low ammonium advantages of N6 medium while optimizing the trace element ratio. By adjusting the concentrations of organic components and premixing sucrose (carbon source) with agar (gelling agent), a solid system more suitable for the subculture and genetic transformation of callus in monocot plants (such as rice and maize) has been developed. This medium is widely used in studies of rice gene editing and maize genetic transformation, particularly in maintaining the embryogenic potential and regenerative capacity of callus, reducing the decline in differentiation ability caused by long-term subculturing.
