WPM Medium（with Sucrose& Agar）

WPM Medium (containing sucrose and agar) is a specialized culture medium developed by Lloyd and McCown in 1980 for woody plant tissue culture. Its core features include low concentrations of inorganic salts (especially very low levels of ammonium nitrogen, with calcium nitrate as the main nitrogen source) and an optimized calcium-to-magnesium ratio, combined with premixed sucrose (as a carbon source) and agar (as a gelling agent), creating a mild solid system suitable for inducing roots, proliferating stem segments, and in vitro preservation of woody plants (such as blueberries, roses, and peach trees). This medium is widely used in the tissue culture of ornamental woody plants and fruit trees, particularly effective in reducing damage to the highly sensitive roots of woody plants caused by high ion concentrations, significantly improving rooting rates and seedling transplant survival.
