2×Quick Clone Mix
Quick Clone is an isothermal seamless cloning enzyme system, which contains T5 exonuclease, high-fidelity DNA polymerase, and Taq DNA ligase. T5 exonuclease possesses strong 5' → 3' exonuclease activity, digesting the cloning fragments to generate 3' overhanging sticky ends. The homologous ends then undergo base pairing, after which the high-fidelity DNA polymerase fills in the gaps in the 5' → 3' direction. Taq DNA ligase connects the 5' phosphate and 3' hydroxyl groups at the nicks to form phosphodiester bonds. Using Quick Clone assembly, one or multiple insert fragments can be directionally, quickly, and accurately inserted into a linearized vector, resulting in a recombinant clone without any extra base sequences. Quick Clone is not limited by restriction sites; linearized vectors can be prepared either by restriction enzyme digestion or directly by PCR. The 5' ends of the PCR primers for the insert fragment are designed to include 15–25 bp sequences identical to the ends of the vector cloning site, serving as homologous arms. The insert fragment with homologous arms and the linearized vector are mixed in a certain ratio, and under the catalysis of recombinase, a 15–20 minute reaction at 50°C is sufficient to allow transformation.
