Mannitol Sodium Chloride Medium/MSB
Mannitol Sodium Chloride Medium/MSB
MSA Medium is a selective and differential medium for staphylococci. It is used to isolate pathogenic staphylococci, such as Staphylococcus aureus.MSA Medium contains an extremely high concentration of sodium chloride. Except for staphylococci in the test sample, the high-concentration sodium chloride inhibits the growth of other microorganisms. Beef extract and polypeptone provide nitrogen sources, carbon sources, sulfur sources, and growth factors for the growth of staphylococci. Phenol red serves as a pH indicator: the phenol red solution is yellow when the pH is < 6.8, red when the pH ranges from 6.8 to 8.4, and fuchsin (fuchsia) when the pH is > 8.4.Staphylococcus aureus ferments mannitol to produce acid, which turns the medium around its colonies yellow. For staphylococci that cannot ferment mannitol (e.g., Staphylococcus epidermidis), the medium around their colonies remains purple or red. Non-pathogenic staphylococci grow less well than Staphylococcus aureus, and their colonies are relatively smaller.
