The core principle of gel recovery is based on the property of silica membranes to specifically adsorb DNA under high salt and low pH conditions, and to release DNA under low salt, neutral or slightly alkaline conditions. The high concentration of ions in the sol solution dissolves the agarose gel, while also shielding the negative charge of DNA and disrupting its hydration layer, exposing the hydrophobic backbone and allowing it to bind firmly to the silica membrane through hydrophobic interactions, hydrogen bonds, and cation bridging; impurities are washed away. After washing, a low ionic strength, slightly alkaline elution buffer removes the cations that shield the DNA charges, restoring its strong negative charge, which leads to electrostatic repulsion and weakens hydrophobic interactions, thereby causing DNA to dissociate from the membrane and dissolve into the buffer, achieving purification.
