1/2MT Medium (containing sucrose and agar) is a low-nutrient derivative formulation of MT medium. Its core feature is reducing the concentrations of macronutrients, trace elements, and organic components in the MT medium to half of the original formula (the iron source is usually maintained at the MT level to avoid iron deficiency), while pre-mixing sucrose (carbon source) and agar (gelling agent) at suitable concentrations, creating a "low-nutrient stress, mild environment" solid system. This medium is widely used in the later stages of plant anther culture for robust seedling growth, somatic embryo germination, and the acclimatization of haploid seedlings in nutrient-sensitive crops such as tobacco and rice. It is particularly effective in balancing seedlings' autonomous metabolic capacity and growth stability under low-nutrient conditions, reducing excessive elongation or malformation caused by high nutrients, and laying a solid foundation for subsequent transplantation.
