DKW Medium(with Sucrose& Agar)
DKW Medium(with Sucrose& Agar)
DKW Medium (containing sucrose and agar) is a specialized woody plant culture medium developed by Driver and Kuniyuki in 1984 for in vitro walnut culture. Its core features include high concentrations of calcium nitrate, a moderate phosphate-to-potassium ratio, and abundant organic nutrients (such as inositol and B vitamins). By optimizing the balance of inorganic salts and pre-mixing sucrose (carbon source) with agar (solidifying agent), it provides a solid system suitable for shoot proliferation, root induction, and in vitro preservation of woody fruit trees like walnut, chestnut, and cherry, as well as hardwood ornamental plants such as rose and rhododendron. This medium is widely used in woody plant tissue culture, particularly meeting the high-calcium requirements of hardwood plants, enhancing root development and stem lignification, and significantly improving the rooting rate and stress resistance of difficult-to-root woody plant seedlings.
