MS Medium(with Sucrose)
MS Medium is a classic general-purpose plant tissue culture medium developed by Murashige and Skoog in 1962. Its core features are a high concentration of inorganic salts (rich in both macronutrients and micronutrients, with nitrogen mainly supplied by ammonium nitrate and potassium nitrate) and a comprehensive combination of organic nutrients. By pre-mixing sucrose (as a carbon source and osmotic regulator, commonly at a concentration of 20-30 g/L), it establishes a liquid or semi-solid culture system suitable for inducing callus formation, shoot differentiation, and rapid proliferation in the vast majority of plants (including monocotyledons, dicotyledonous herbaceous plants, and some woody plants) (without added gelling agents, allowing for either liquid culture or subsequent agar addition as needed). This medium is the most widely used in plant tissue culture, particularly providing ample nutrients for plant explants, supporting rapid growth and proliferation, and serving as the basic system for tissue culture and genetic transformation experiments in crops, ornamental plants, and medicinal plants.
