1/4MS Medium is a ultra-low concentration derivative of the MS medium, primarily achieved by reducing the concentration of macronutrients in the MS medium to one-quarter of the original formula. Trace elements, organic components, and iron sources are usually kept at full MS levels, while sucrose is added as a carbon source and osmotic regulator, creating a culture system of 'extremely low nutrient supply and mild ionic stress' (agar is not added; liquid culture or solid medium with gelling agents can be chosen according to needs). This medium is mainly suitable for the culture of plant materials sensitive to high ion concentrations, the acclimation stage of tissue-cultured seedlings transitioning from higher nutrient environments to natural conditions, and the dedifferentiation and growth of certain special crop tissues. It can reduce ionic toxicity through a gentle nutrient environment and help plant materials adapt to low-nutrient conditions.
