3/4 MS Medium（with Sucrose）

3/4MS medium is a medium-concentration derivative of MS medium. Its key feature is that the concentrations of macronutrients, micronutrients, and organic components in MS medium are adjusted to three-quarters of the original formula (with the iron source usually maintained at MS level), and sucrose (typically 25 g/L) is pre-mixed as a carbon source and osmotic regulator. This creates a liquid or semi-solid culture system with 'moderate to high nutrient supply and moderate osmotic pressure' (without agar; agar can be added as needed for solid culture). This medium is widely used in plant tissue culture for seedling growth, secondary shoot induction, and cultivation of moderately sensitive materials. It particularly balances nutrient supply with environmental adaptability, filling the nutritional gradient gap between MS and 1/2MS, making it suitable as an intermediate culture for transitioning from rapid proliferation to rooting stages.
