B5 Medium is a classic plant tissue culture medium. Its core characteristics are a high content of potassium nitrate and a lower proportion of ammonium nitrogen in the inorganic salts, combined with a high concentration of vitamin B1. Sucrose is added as a carbon source and osmotic regulator, creating a culture system suitable for dicotyledonous plants and some sensitive materials (without agar, it can be flexibly used for liquid culture or later solidified to make solid media). This medium is widely used for callus induction and proliferation in dicotyledonous plants, protoplast culture, and genetic transformation. It is particularly effective in reducing the toxicity of ammonium nitrogen to sensitive materials, promoting cell division and callus growth, and is suitable for plant tissue culture scenarios that require a specific nitrogen-to-potassium ratio.
