N6 Medium is a classic culture medium specifically designed for the in vitro cultivation of gramineous crops. Its key features are a low ammonium nitrogen and high potassium inorganic salt ratio, combined with sucrose as a carbon source and osmotic regulator, creating a culture system tailored to the growth requirements of gramineous plants (agar is not added; liquid culture can be chosen or a gelling agent can be added to make solid medium as needed). This medium was initially used for rice anther culture and is now widely applied to callus induction, embryoid formation, and genetic transformation in gramineous crops such as rice, wheat, and maize. It helps reduce the adverse effects of high ammonium on excised plant tissues, providing a suitable nutritional environment for their growth and differentiation.
