De Man, Rogosa and Sharpe Agar Medium

MRS medium is used for the enrichment, culture, and isolation of lactic acid bacteria according to the method of DeMan, Rogosa, and Sharpe. It can also be used for the identification and testing of lactobacilli. Peptone provides a nitrogen source, glucose provides a carbon source, beef extract and yeast extract serve as comprehensive sources of nutrients and vitamins, while sodium acetate and ammonium citrate act as selective agents and energy sources. Sodium acetate inhibits Gram-negative bacteria and fungi but has no effect on lactic acid bacteria. Magnesium sulfate and manganese sulfate provide essential ions for metabolism. Tween 80 is a non-ionic surfactant that helps lactobacilli utilize nutrients. The slightly acidic environment of the medium is conducive to the growth of lactic acid bacteria.
