NS Medium is a specialized medium commonly used in the fields of plant protoplast culture and somatic hybridization. Its core characteristics are the low-osmotic inorganic salt composition and an optimized combination of organic nutrients, with added sucrose serving as both a carbon source and an osmotic regulator, creating a gentle culture system (without added agar, often used in liquid culture to suit protoplast growth requirements, although a gelling agent can be added as needed). This medium is primarily suitable for the survival, cell wall regeneration, and callus formation of protoplasts from crops such as tobacco and tomato. It helps reduce osmotic stress from high ion concentrations in cell wall–free protoplasts, thereby facilitating subsequent protoplast division and development.
