1/2 MT Medium is a reduced-nutrient derivative of MT medium. Its core principle is to lower the concentrations of macronutrients, micronutrients, and organic components in MT medium to half of the original formulation (the iron source is usually maintained at MT levels to avoid iron deficiency), while adding sucrose as a carbon source and osmotic regulator to create a "low-nutrient stress, mild environment" cultivation system (agar is not added, so it can be used as liquid medium or solidified with gelling agents as needed). This medium is mainly used for the later-stage seedling growth of plant anther cultures, somatic embryo germination, and the acclimatization of haploid seedlings in nutrient-sensitive crops. It helps balance autonomous metabolism and growth stability in seedlings under low-nutrient conditions, reducing overgrowth or deformities caused by high nutrient levels, thereby laying a strong foundation for subsequent transplantation.
