PMSF(100×)
PMSF(100×)
PMSF exerts its inhibitory effect by sulfonating the hydroxyl group of the serine residue at the active site of serine proteases. At the same time, PMSF can also inhibit papain, which belongs to cysteine proteases, and acetylcholinesterase in mammals, but it does not inhibit metalloproteases, most cysteine proteases, or aspartic proteases. The effective concentration of PMSF is 0.1–1 mM. PMSF is a low-toxicity alternative to diisopropylfluorophosphate (DFP).PMSF is unstable in water. Under certain conditions, such as alkaline pH, its half-life may be only 35 minutes. It is recommended to add PMSF to lysis buffer or other aqueous solutions just before use.
