Proteinase K solution can be used to digest various proteins. It is used in a variety of common molecular biology, cell biology, and related experiments, such as genomic DNA extraction and enzymatic digestion removal. Enzyme activity is >30 U/mg. At 37°C, using hemoglobin as a substrate, the amount of proteinase K that produces an equivalent of 1 μmol of tyrosine Folin-positive amino acids or peptides within 1 minute is defined as one unit of proteinase K activity. It is effective over a wide pH range, with an effective pH range of 4.0-12.5 and an optimal pH range of 7.5-8.0. The optimal reaction temperature for proteinase K is 65°C, but at 65°C or higher, proteinase K itself degrades very rapidly. Often, a reaction temperature of 50-55°C is chosen. Proteinase K shows higher enzyme activity in the presence of 0.2-1% SDS or approximately 10 mM urea. The commonly used working concentration of proteinase K is 0.05-1 mg/ml, depending on factors such as whether the buffer contains SDS or urea, whether the pH is suitable, and whether the temperature is appropriate. Common concentrations of EDTA, Triton X-100, Tween 20, Sarkosyl, and guanidine hydrochloride have little effect on the activity of proteinase K.
