IPTG
IPTG is an inducer of β-galactosidase activity. In blue-white screening, IPTG serves as a gratuitous inducer to stimulate protein expression in bacteria, while X-gal acts as a chromogenic substrate. The principle is that IPTG can induce the Lac operon segment of the vector to synthesize the N-terminal fragment of β-galactosidase, which can complement the defective β-galactosidase encoded by the host cell (α-complementation). Bacteria with α-complementation grown on a medium containing the X-gal chromogenic substrate form blue colonies. However, if exogenous DNA is inserted into the plasmid's multiple cloning site, it disrupts the α-complementation, resulting in white colonies on the medium. IPTG is also commonly used as an inducer for recombinant protein expression in genetic engineering.
