IPTG Dioxane Free
IPTG is an inducer of β-galactosidase activity. In blue-white screening, IPTG acts as a gratuitous inducer to trigger protein expression in bacteria, while X-gal serves as a chromogenic substrate. The principle is that IPTG induces the Lac operon DNA segment of the vector to synthesize the amino-terminal fragment of β-galactosidase, which can complement the defective β-galactosidase encoded by the host cell (α-complementation). Bacteria exhibiting α-complementation, when plated on media containing the chromogenic substrate X-gal, form blue colonies. However, if foreign DNA is inserted into the multiple cloning site of the plasmid, it can disrupt α-complementation, and white colonies will grow on the medium. IPTG is also a commonly used inducer for recombinant protein expression in genetic engineering.
