BU Buffer, also known as BU Salt, is a reagent used in yeast hybrid systems such as EGY48 and NMY51 to detect the Lacz reporter gene. When an interaction occurs, the LacZ reporter gene is activated, producing β-galactosidase, which cleaves X-gal into galactose and the deep blue substrate 5-bromo-4-chloro-indolyl, thereby making positive colonies appear blue.
