Imidazole
Imidazole
Imidazole is a five-membered heterocyclic compound found in many natural compounds. It exhibits both acidic and basic properties. It has been reported to be an inhibitor of thrombosis. Imidazole is slightly more basic than pyrazole and can form stable salts, such as bitter salts and nitrates. One application of imidazole is in immobilized metal affinity chromatography (IMAC) for the purification of His-tagged proteins. The tagged proteins bind to nickel ion media within the pores of the chromatographic column, and excess imidazole is passed through the column to elute the nickel-coordinated tagged proteins, yielding highly pure target proteins. It is especially suitable for buffers in the pH range of 6.2 to 7.8.
