R2A Liquid Medium
R2A, also known as Medium S, was developed by Reasoner and Geldreich. R2A is a low-nutrient medium used for the microbiological plate count of treated drinking water. The medium allows for the recovery of damaged and chlorine-tolerant bacteria without being overshadowed by rapidly growing microorganisms. R2A achieves this requirement by combining extended incubation times and low incubation temperatures (optional), yielding significantly higher recovery rates for fatigued microbes. Acid-hydrolyzed casein and peptone provide nitrogen, vitamins, amino acids, carbon, and minerals. Yeast extract provides vitamins and essential trace elements; soluble starch can adsorb toxic metabolic byproducts, benefiting the recovery of damaged microorganisms; sodium pyruvate aids the recovery of stressed cells; magnesium sulfate provides divalent cations and sulfur; and potassium phosphate is a phosphorus source that can also buffer the pH of the medium.
