Malt Extract Agar Medium (MEA)
Malt Extract Agar (MEA) is a liquid enrichment medium for yeast and mold, recommended for the detection, isolation, and counting of fungi in various materials, particularly yeasts and molds. There are several improved versions of the Malt Extract Agar formula, primarily recommended for sterile testing and also for cultivating strains used in the vitamin microbial assay. The nutritional requirements of yeasts and molds are lower than those of bacteria, allowing them to grow on media with lower nutrient content. Additionally, molds and yeasts can adapt to a wide pH range, being able to proliferate under pH 3.4 conditions, while the vast majority of bacteria cannot grow. Malt Extract Agar utilizes these two characteristics to selectively isolate and enrich molds and yeasts. Adding acid-stable antibacterial agents, such as aureoflavin, to Malt Extract Agar can further inhibit bacterial growth. The maltose, dextrin, and glycerol in the Malt Extract Agar formula serve as carbon sources and energy substrates to meet the needs of different microbial species, while peptone acts as an organic nitrogen source.
