Kanamycin sulfate is an aminoglycoside antibiotic that inhibits Gram-positive and Gram-negative bacteria as well as mycoplasma. Its mechanism of action involves specifically binding to the bacterial 70S ribosomal subunit, thereby inhibiting ribosomal translocation and causing misreading, which further disrupts protein synthesis. Kanamycin sulfate can be used in cell culture to prevent bacterial contamination. In molecular biology research, it is commonly used to selectively screen bacterial clones that have successfully been transformed with the kanamycin resistance gene (kan gene); it is also frequently used in Agrobacterium-mediated transformation experiments to selectively screen plant tissues carrying the nptII (APH3) gene.
