Carrier DNA (salmon sperm DNA) is short linear single-stranded DNA that can encapsulate plasmid DNA and plays a role in the uptake of exogenous plasmid DNA by yeast cells. It is an indispensable component in yeast transformation experiments, effectively promoting plasmid entry into yeast cells and improving transformation efficiency. Additionally, it can protect plasmids from degradation by DNase. Carrier DNA must be heat-denatured before each use to ensure that it exists in single-stranded form in the transformation system. The concentration of the Carrier DNA solution is 10 mg/ml, and only 5 μl of Carrier DNA is needed for each transformation reaction.
