Spectinomycin hydrochloride, an aminocyclitol antibiotic, has a strong inhibitory effect on both Gram-positive and Gram-negative bacteria. Its antibacterial mechanism involves binding to the 30S subunit of bacterial ribosomes, inhibiting protein synthesis and elongation by interfering with the translocation of peptidyl-tRNA. Mutations in the ribosomal protein 5S gene rpsE (ribosomal protein S5 gene) can prevent the binding of spectinomycin hydrochloride, resulting in resistance to this antibiotic.
