G418 (Geneticin) stock solution (50 mg/ml sterile)
G418 (Geneticin) is an aminoglycoside antibiotic commonly used as a selective agent for stable transfection in molecular genetics experiments. It inhibits the genes of transposons Tn601 and Tn5, interferes with ribosome function, and blocks protein synthesis, producing toxic effects on both prokaryotic and eukaryotic cells, including bacteria, yeast, plants, mammalian cells, as well as protozoa and worms. When the neo gene is integrated into the DNA of eukaryotic cells, it can initiate transcription of the neo gene-encoded sequence into mRNA, leading to efficient expression of the resistance protein aminoglycoside phosphotransferase. This enables the cells to acquire resistance and grow in selective media containing G418. This selective property of G418 has been widely applied in gene transfer, gene knockout, resistance screening, and the generation of transgenic animals.
