2×YPDA Medium

2×YPDA provides twice the nutrition of regular YPDA and can improve mating efficiency during the formation of zygotes from different yeast gametes. YPDA medium is a nutrient-rich yeast medium, also known as a complete medium, and is non-selective. YPDA medium is composed of (Y) yeast extract, (P) peptone, (D) dextrose (glucose), and (A) adenine. Yeast extract and peptone provide essential nitrogen sources, vitamins, and growth factors; glucose supplies a carbon source; and adenine sulfate can prevent yeast strains with ADE1 and ADE2 mutant alleles (such as Y2HGold, AH109, and Y187) from turning pink or red after extended culturing.
