0.5×YPDA liquid medium

0.5×YPDA provides half the nutrients of regular YPDA. After low-speed centrifugation of the cells at the end of the mating experiment, resuspending yeast in 0.5×YPDA liquid medium and plating can achieve higher mating efficiency compared to resuspension in 0.9% (w/v) NaCl or ddH2O. YPDA medium is a nutrient-rich medium for yeast, also known as a complete medium, and it is non-selective. YPDA medium is composed of (Y) yeast extract, (P) peptone, (D) dextrose (glucose), and (A) adenine. Yeast extract and peptone provide essential nitrogen sources, vitamins, and growth factors; glucose provides a carbon source; and adenine sulfate prevents yeast strains with ADE1 and ADE2 mutant alleles (such as Y2HGold, AH109, and Y187) from turning pink or red after long-term culture.
