Glufosinate (PPT)

Glutamine synthetase plays a role in plant nitrogen metabolism. It is an important detoxifying enzyme in plants that can neutralize the toxicity of ammonium released during nitrate reduction, amino acid degradation, and photorespiration. The target enzyme of glufosinate is glutamine synthetase (GS). Under normal conditions, GS can form λ-glutamyl phosphate from ATP and glutamate. However, after PPT treatment, PPT first binds with ATP, and the phosphorylated PPT occupies the eight active sites of the GS molecule, causing a change in the spatial configuration of GS, which in turn inhibits GS activity. There are two forms of GS in plant leaves: GS1 is present in the cytoplasm, and GS2 is present in the chloroplast. The activity ratio of the two forms also varies among different plant leaves, but PPT can inhibit all known forms of GS. As a result of GS inhibition by glufosinate, nitrogen metabolism in the plant is disrupted, ammonium accumulates excessively, chloroplasts disintegrate, photosynthesis is inhibited, and ultimately the plant dies.
