G418 (Geneticin)
G418 (geneticin) is an aminoglycoside antibiotic that is commonly used as a selection reagent for stable transfection in molecular genetics experiments. It blocks protein synthesis by inhibiting the genes of transposons Tn601 and Tn5, thereby interfering with ribosome function, and exerts toxic effects on both prokaryotic and eukaryotic cells, including bacteria, yeast, plants, mammalian cells, as well as protozoa and worms. When the neo gene is integrated into the DNA of eukaryotic cells, it can initiate the transcription of the sequence encoded by the neo gene into mRNA, leading to the efficient expression of the resistance product aminoglycoside phosphotransferase. This enables cells to acquire resistance and grow in selective media containing G418. This selective property of G418 has been widely applied in gene transfer, gene knockout, resistance screening, and transgenic animals.
