5-Fluoroorotic Acid(5-FOA)
5-Fluoroorotic Acid(5-FOA)
5-Fluoroorctic acid (5-FOA) is a commonly used molecular genetic screening method for yeast negative selection. 5-FOA is employed in yeast plasmid clearance, plasmid shuttle, positional exchange, and double-hybrid screening. 5-FOA itself is non-toxic to yeast cells, but the functional URA3 gene encodes and expresses orotic acid nucleoside-5'-monophosphate decarboxylase (OMP decarboxylase), which converts it into toxic forms (e.g., 5-fluorouracil), thereby inhibiting yeast cell growth. Consequently, in 5-FOA media supplemented with uracil, yeast strains carrying the URA3 mutant gene can grow normally. 5-FOA is primarily used for the identification and screening of Saccharomyces cerevisiae (URA3), although it has also been applied to other strains, including Schizosaccharomyces pombe (URA4 and URA5), Candida albicans (URA3), and Escherichia coli (pyrF). The use of 5-FOA in yeast double-hybrid systems enables the construction of activation domain hybridization libraries for the identification of protein-protein interactions.
