Terrific Broth (TB), also known as TB medium, is an enriched medium derived from the improvement of LB medium. TB medium is commonly used as a growth medium for E. coli in molecular biology experiments, with its most notable feature being the ability to achieve high cell density. Glycerol serves as the energy source in this medium; since glycerol fermentation does not produce acid, it does not rapidly lower the pH of the culture. Phosphates provide a buffering system to maintain pH stability and prevent cell death caused by a drop in pH. Casein hydrolysate supplies nitrogen, vitamins, minerals, and amino acids, while yeast extract is a rich source of vitamins. TB medium is mainly used for recombinant protein expression in E. coli, yielding high amounts of highly soluble proteins. It is suitable for various protein expression methods, such as IPTG induction, arabinose induction, and maltose induction. Compared with LB medium, using TB medium can increase plasmid DNA yield by 2–4 times. Super Broth (SB), abbreviated as SB medium, is richer in nutrients compared to TB medium. Casein hydrolysate and yeast extract provide nitrogen sources, vitamins, and other essential nutrients for growth, while sodium chloride maintains osmotic balance. It is suitable for non-selective cultivation of lambda phage and for large-scale cultivation of E. coli and recombinant protein expression. With the addition of antibiotics, it can be used for selective cultivation.
