Chapman agar
Chapman Agar is a highly selective identification medium, which is mainly used for the separation of staphylococcus, especially for the identification of pathogenic staphylococcus aureus. Its formula contains significantly high concentration of sodium chloride (up to 7.5%), which can effectively inhibit the growth of most gram-negative bacteria and gram-positive cocci, while staphylococcus can tolerate it. Mannitol in the culture medium is used as fermentable sugar and phenol red is used as acid-base indicator. When pathogenic Staphylococcus aureus ferments mannitol to produce acid, the culture medium around the colony will change from red to bright yellow. This characteristic can distinguish it from coagulase-negative staphylococcus which does not ferment mannitol, and it is a classic method to identify Staphylococcus aureus in clinical and food detection.
