XLD agar
XLD agar (xylose-lysine-deoxycholate agar) is a highly selective and differential medium, which is specially used for the separation and identification of Salmonella and Shigella. Its selectivity depends on deoxycholate and ferric ammonium citrate, and it can inhibit gram-positive bacteria and some coliforms. The identification system is based on xylose, lactose, sucrose and lysine decarboxylase reactions. Salmonella (lysine decarboxylase positive) ferments xylose to produce acid, which turns the colony yellow, and then uses lysine to produce alkali to restore red. The typical colony is red with black center (due to hydrogen sulfide production). Shigella (lactose-free, lysine-negative) is a red transparent colony. Bacteria that ferment lactose/sucrose (such as Escherichia coli) form opaque yellow colonies. This feature makes the target pathogen easy to identify.
