Magnesium chloride malachite green broth (MM,RV)
Magnesium chloride malachite green broth (MM,RV)
Magnesium chloride malachite green broth (Rappaport-Vassiliadis broth, RV broth for short) is a highly selective enrichment solution, which is specially used for the proliferation of Salmonella in food and environmental samples. Its strong selectivity is based on a unique combination of inhibitors: high concentration of magnesium chloride creates osmotic stress, while malachite green inhibits the growth of most non-Salmonella (including Escherichia coli and Proteus). This dual pressure provides a competitive advantage for Salmonella. The medium was used at a relatively low culture temperature (42°C) to further enhance the selectivity. Inoculating the pre-enriched samples into this broth can make a small amount of Salmonella proliferate rapidly, significantly improving the sensitivity of subsequent detection on selective plates, which is the core step of modern standard methods for Salmonella detection.
