Bacterial Two-hybrid Kit
Using the catalytic domain (CyaA) of adenylate cyclase from Bordetella pertussis, which consists of two complementary fragments, T25 and T18, that are inactive when physically separated. By fusing these two fragments to interacting peptides X and Y, if X and Y interact, it leads to functional complementation between the T25 and T18 fragments, thereby promoting the synthesis of cAMP. cAMP binds to catabolite activator protein (CAP), which in turn activates the expression of genes including the lac and mal operons, allowing bacteria to utilize lactose or maltose as a carbon source and making them easily distinguishable on indicator or selective media.
