V-P semisolid agar

V-P semisolid agar is a special culture medium for detecting V-P reaction of microorganisms, which is mainly used to identify Escherichia coli and Enterobacter aerogenes. Its basic components provide necessary nitrogen source, carbon source and growth factor, while the key components are glucose (or sodium pyruvate) and buffer system. Microorganisms metabolize glucose to produce the precursor of acetyl methyl methanol (3- hydroxy -2- butanone), which is oxidized to diacetyl in alkaline environment, and the latter reacts with guanidine group of arginine in the culture medium to produce red compounds. The semi-solid form is not only beneficial to the growth of facultative anaerobic bacteria, but also convenient to observe the diffusive reaction. Usually, potassium hydroxide and α -naphthol reagents are added after culture, and a red ring appears on the liquid surface, which is a positive reaction. This culture medium is of great significance to the identification of intestinal flora.
