The cDNA normalization kit uses a cDNA normalization method based on duplex-specific nuclease (DSN). This method relies on nucleic acid hybridization kinetics and the unique properties of DSN. Due to the thermal stability of DSN, the degradation of dsDNA occurs under cDNA reannealing conditions, which prevents the formation of secondary structures and nonspecific hybridization involving intramolecular sequences in single-stranded (ss) cDNA fragments. During reannealing, high-abundance genes are more efficiently converted into double-stranded cDNA. This clearly forms two fractions: a high-abundance cDNA portion and normalized single-stranded cDNA. Subsequently, the double-stranded cDNA is degraded by DSN, and the remaining normalized single-stranded cDNA is amplified via PCR.
