Glucose semisolid medium
Glucose semi-solid medium is a multifunctional identification medium which integrates the observation of microbial motility and the preliminary judgment of sugar fermentation characteristics. Its basic components are peptone, beef paste and other nitrogen sources and growth factors, and a small amount (about 0.3%-0.5%) of agar is added to make semi-solid state, as well as glucose and phenol red indicators. Puncture method was used during inoculation. If bacteria can move, they will spread and grow around along the puncture line, making the whole culture medium turbid; If you can't move, you will only grow along the puncture line. At the same time, if bacteria can ferment glucose to produce acid, phenol red indicator turns yellow; If gas is produced, bubbles or fractures will appear in the semi-solid. After puncture inoculation, the culture medium can observe three indexes of motility, acid production and gas production by sugar fermentation at one time, which is widely used in the preliminary identification of Enterobacteriaceae and other bacteria, with high efficiency and simple operation.
