Plant transcription activity detection kit

The dual-luciferase (Dual-Luc) reporter gene system for detecting plant transcriptional activity can be applied to analyze the transcriptional activity of plant proteins. Firefly luciferase (FLuc) and Renilla luciferase (RLuc) are co-expressed in the same cell, and each can catalyze the oxidation of luciferin and coelenterazine, respectively, producing luminescence, which can be detected using a fluorescence detector. RLuc serves as an internal reference gene to reduce experimental errors caused by external factors such as cell viability and transfection efficiency. GAL4BD serves as the basal expression, and GAL4BD-VP16 can be used as a positive control for transcriptional activation. The transcription factor to be tested is fused with GAL4 BD, and BD binds to the UAS transcriptional regulatory element to regulate FLuc expression. The ratio of FLuc to RLuc can be used to determine whether the transcription factor has transcriptional activation or repression activity.
