Lead acetate medium
Lead acetate medium is a biochemical identification medium specially used to detect the ability of microorganisms to produce hydrogen sulfide (H2S). Its basic components are usually peptone and beef extract powder, which provide nitrogen source, carbon source and growth factor, and sulfur-containing amino acids (such as cystine) as sulfur source. The key component is lead acetate, which is used as an indicator of H2S. If bacteria can decompose sulfur-containing amino acids to produce H2S, the gas reacts with lead ions in the culture medium to produce insoluble black lead sulfide precipitate. Observing this black precipitate can be judged as a positive reaction. This culture medium is often used to identify bacteria such as Enterobacteriaceae, such as Salmonella (usually positive) and Escherichia coli (negative). Because of its high sensitivity and intuitive interpretation, it is a classic method in microbiological examination.
